*Well I dont have the V6 but based on previous discussions my understanding is that its located underneath the intake. Not sure if its between the upper and lower or all the way down...but everyone says its a biatch to get at. Check your wire as far as you can first and maybe save yourself some work. Are you getting an error code related to the knock sensor? Generally they dont go bad...but the wire commonly does.

man its NOT easy. pretty much at the center of the motor, you pretty much tear down the whole top end to get to it.

It's not too hard. You WILL also want to replace the wire that runs between the head and intake manifold. After paying the cost on the knock sensor and intake manifold gaskets, the wire is cheap.

Plan on extra expenses. Hoses you might not know existed may need replaced. You may want to replace your timing belt and check both your idler pulleys since you'll be in the area anyway.

Body Damage is Cool
Toyota replaced my truck's knock sensor wire at no cost to me when they did my headgasket. They claim these wires have given them trouble due to heat, so you might want to look there first.

You are looking in the wrong place. The wire is under your throttle body.

Here's a picture I stole:
